Proinflammatory response was associated with reduced intracellular zinc and was enhanced by zinc deficiency. THP-1 cells (n=3 per treatment) were cultured in ZA or ZD media for 10 to 13 days. Total zinc (A) and intracellular zinc (B) were determined by ICP-OES and FluoZin-3 flow cytometry, respectively. Intracellular zinc and proinflammatory response were measured in ZA or ZD THP-1 cells that were left untreated (UT), or stimulated with 10 or 100 ng/ml LPS for 6 h. Expressions of TNFα (C) and IL1β mRNA (D) were determined by real-time PCR. Data represent mean±S.E.M. MFI, mean fluorescence intensity. *P<.05 vs. ZA.
